It sounds like your A500 might be having some hardware compatibility issues rather than just a software limitation. While most non-AGA games should work on the A500, some later ECS titles were optimized for the A600/A1200 and might struggle on the older chipset. Have you checked if your Kickstart version is causing conflicts? Also, since WinUAE runs the game fine, it could be a RAM or drive issue on your real hardware. Maybe try cleaning the floppy drive heads or using another Kickstart ROM if possible!
